• It’s hypothesized that the cleavage of the endoperoxide bridge within the pharmacophore of DHA generates reactive oxygen species (ROS), which increases oxidative stress and causes malarial protein damage via alkylation.
• Sulfadoxine is a sulfonamide antibiotic that competes with p-aminobenzoic acid within the biosynthesis of folate. Pyrimethamine is a selective inhibitor of protozoal dihydrofolate reductase, hence preventing the synthesis of tetrahydrofolate — the active sort of folate.
